Routine Examination of Urine

Virtual practical for routine examination of urine with emphasis on bacteriology and parasitology.

The practical includes physical examination, chemical examination, urinary sediment microscopy, bacteriological culture, incubation, Gram staining and parasitological examination.

Simulation timing: Centrifugation takes exactly 5 seconds. Incubation takes exactly 30 seconds.

Practical Observations

Physical examination Yellow to amber urine, normally clear to slightly hazy. Specific gravity commonly approximately 1.003–1.030 depending on hydration and renal concentrating ability.
Chemical examination Record pH and reagent-strip reactions for protein, glucose, ketones, blood, bilirubin, urobilinogen, nitrite and leukocyte esterase.
Microscopic sediment Examine RBCs, WBCs, epithelial cells, bacteria, yeast, crystals, casts and other formed elements.
Bacteriology Interpret bacteriuria using colony count, specimen quality, pure or mixed growth, colony morphology, Gram reaction, identification and susceptibility testing.
Parasitology Concentrated urine sediment may be examined for parasites. Schistosoma haematobium eggs characteristically possess a prominent terminal spine.
Interpretation Findings must be interpreted together with specimen quality, clinical information and the possibility of contamination.

Routine Urine Examination — Final Report

Examination Observation / Finding Interpretation
Colour Yellow Within the usual normal range.
Appearance Clear to slightly hazy No gross abnormal turbidity in this simulated specimen.
Specific gravity Approximately 1.015 Compatible with a normally concentrated specimen.
pH Approximately 6 Slightly acidic urine.
Protein Negative No detectable proteinuria in this simulated specimen.
Glucose Negative No detectable glucosuria.
Ketones Negative No detectable ketonuria.
Blood Negative No significant hematuria/ hemoglobin reaction.
Nitrite Negative No nitrite reaction in this simulated specimen.
Leukocyte esterase Negative No significant leukocyte esterase reaction.
Microscopy Occasional epithelial cells; no significant RBC/WBC excess No marked inflammatory sediment in this simulated specimen.
Bacterial culture No significant pure bacterial growth in this simulation Correlate with clinical findings and specimen quality.
Parasitology No parasite detected No demonstrable urinary parasite in this simulated specimen.
Important interpretation point: In actual laboratory practice, urine microscopy, culture and susceptibility testing should not be interpreted independently. Bacterial culture results should be assessed together with colony count, specimen type, collection method, contamination risk, symptoms and the organism recovered.
